Lens Configurations

The V4 Miniscope's working distance (distance between the bottom of the Miniscope and focal plane) and Field-of-View (FOV) can be adjusted based on the combination of lenses used when assembling the Miniscope. Below lists suggested lens combinations with their resulting optical properties.

Refer to the below picture when this guide refers to lens "1", lens "2", and lens "3". Lenses "1" and "2" comprise the Objective while lens "3" acts as the achromatic tube lens.

Lens Configuration 1

This is what we consider the standard V4 Minsicope lens configuration. It gives a good balance or resolution, FOV, and working distance and will work for deep imaging through rely GRIN lenses as well as cortical imaging through thin cranial windows.

Optical properties:

  • Field-of-View: 1mm x 1mm diameter
  • Working Distance: 675um +/-250um

Lens Configuration 2

This lens configuration extends the working distance slightly which can make imaging through a cranial window a bit easier. It also results in a slightly larger FOV when compared to Lens Configuration 1 with the trade off of a slight reduction in spatial resolution and effective NA (light collection).

Optical properties:

  • Field-of-View: 1.1mm x 1.2mm diameter
  • Working Distance: 975um +/-340um

Lens Configuration 3

This lens configuration significantly extends the working distance as well as the FOV. The resulting system is at the edge of having single cell resolution and collects noticeably less light than Lens Configuration 1.

Optical properties:

  • Field-of-View: 1.3mm x 1.4mm diameter
  • Working Distance: 2010um +/-400um
